和胃汤对大鼠实验性胃黏膜损伤的影响 被引量:4

摘要 目的:探讨和胃汤抗胃黏膜损伤的作用及其机理。方法:将80只大鼠随机分为4组,采用吲哚美辛灌胃建立大鼠实验性胃黏膜损伤模型,观察和胃汤时胃黏膜损伤的影响,并测定血浆一氧化氮(NO)、内皮素(ET-1)含量;测定胃黏膜前列腺素(PG)、生长抑素(SS)含量。结果:肉眼模型对照组可见散在胃黏膜浅糜烂;光镜下模型对照组大鼠胃黏膜有明显损伤,呈暗红色充血、水肿、大量散在出血点、条索状糜烂,以胃体部为主,整个黏膜层细胞有明显变性、坏死或脱落,固有膜有灶性出血及急性炎症反应。和胃汤组、丽珠得乐组胃黏膜可见少量出血点及糜烂,镜下胃黏膜表面有少量炎性渗出物,胃黏膜浅层细胞有变性、坏死。胃黏膜损伤指数模型对照组大鼠最高,和胃汤组、丽珠得乐组较低,与模型对照组比较,差异有非常显著性意义(P<0.01)。血浆NO含量和胃汤组显著升高,与模型对照组、丽珠得乐组比较,差异有非常显著性意义(P<0.01)。ET-1含量和胃汤组与丽珠得乐组均低于模型对照组(P<0.01)。胃黏膜内PG含量模型对照组明显降低,显示胃黏膜有明显损伤。和胃汤组、丽珠得乐组胃黏膜内PG含量较高,与模型对照组比较,差异有非常显著性意义(P<0.01)。和胃汤组、丽珠得乐组与模型对照组比较,胃黏膜内SS含量也明显升高(P<0.05),而和胃汤组、丽珠得乐组之间比较,差异无显著性意义(P>0.05)。结论:和胃汤具有保护胃黏膜的作用,其机理可能与影响体内ET-1、NO、PG、SS水平有关。 Objective:To observe the therapeutic effect of Hewei Decoction(HD)on experimetal gastric mucosal lesion(GML)and to explore its mechaniam.Methods:Eighty rats were randomized into 4 groups.The rat model of experimental gaetric mucosal was induced by gastric gavage of indomethacin.The effect of HD on the GML was evaluated.The changes of plasma nitric oxide(NO)and endothelin(ET-1),and the gastric mucosal prostaglandin(PG)and somatostatin(SS)were observed before and after treatment.Re- sults:Scattered gastric mucosal erosion was found in the model group by naked eyes.Under light microscope,there existed obvious signs d gastric mucosal lesion:dark red conges- tion,swelling,large amount of scattered hemo(?)thagic apots,strip-shaped lesion mainly lo- cated in gsstric beds,cell degeneration,necrosis sad abscission in the whole mucosa lay- er.focal bleeding and acute inflammatory reaction in the lamina propria.While there were less hemorrhagic spots,gastric mucosul erosion,inflammatory exudates,cell degeneration or necrosis in HD group and Bismuth Potassium Citrate Granules group.The gastric muco(?)al le- sion index was the highest in the model group,and higher in HD group and Biamuth Potas- sium Citrate Granules group(P<0.01 compared with that in the model group).The plasma NO content was increased in HD group as compared with that in the model group and Bis- mulh Potassium Citrate Granules group(P<0.01).The ET-1 content was lower but PG content was higher in HD group and Bismuth Potassium Citrate Granules group than those in the model group(P<0.01).Mucosal SS content was also increased in HD group and Bis- muth Potassium Citrate Granules group(P<0.05 compared with that in the model group), but the difference was insignificant between HD group and Bismuth Potassium Citrate Gran- ules group(P>0.05).Conclusion:HD has protective effect on gastric mucoss,and its mechanism may be related with the regulation of ET-1,NO,PC,and SS levels.
