摘要
目的建立HPLC法测定Beagle犬血清中茶碱的浓度,并探讨其药动学过程。方法以咖啡因为内标,用15%高氯酸溶液作为蛋白沉淀剂,以甲醇-1%醋酸-三乙胺(24:76:0.15,V/V)为流动相,以岛津Skim-pack VP-ODS C_(18)(150 mm×4.6 mm,5μm)为色谱柱;柱温25℃;流速1 mL·min^(-1);检测波长(275±2)nm;测定Beagle犬血清中茶碱的浓度。结果茶碱血清线性范围为0.1~25 mg·L^(-1),检测限为25μg·L^(-1),方法回收率均>95%,日内、日间精密度均符合要求。结论此方法可用于茶碱血药浓度分析及雾化吸入氨茶碱后的药动学研究。
AIM To develop a HPLC method for the detemfination of theophylline in serum and to investigate its pharmaeokinetic eharaeteristies in Beagle's. METHODS Serum samples (using eaffeine as internal standard) were sedimented by 15% perehlorie acid and then detected by UV detector at (275 ± 2)nm with Shim-paek VP-ODS C18( 150 mm × 4.6 mm, 5 μm ) and CH3OH: CH3COOH( 1% ) : trolamine solution (24:76:0.15) as mobile phase at a flow rate of 1 mL·min^-1 the serum eoneentration was deteeted by HPLC with the eolumn temperature of 25℃. RESULTS The linear range was 0.1 - 25 mg·L^- 1. The detectable eoneentration was 25μg·L^- 1. The method recovery was more than 95%. Both the inter-and intra-day precision (RSD) was less than 5.0%. CONCLUSION The method provides a stable and reliable method for pharmaeokinetic studies after inhaling aminophylline as well as detemfining eoneentration of theophylline in serum.
