摘要
目的观察基质细胞衍生因子-1α(SDF-1α)在颈总动脉球囊损伤后血管中的表达。方法60只雄性SD大鼠分成对照组(C组)和球囊损伤组(S组),后者又分为术后即刻组(S0组)和术后1 d、4 d、7 d组(S1、S4、S7组)。S组大鼠均行左侧颈总动脉球囊损伤术。各组大鼠分别采血,流式细胞仪检测外周血中CD34+CXCR4+细胞;取左侧颈总动脉,应用RT-PCR技术和Western blotting法分别检测SDF-1αmRNA和蛋白表达。结果S0、S1、S4组大鼠外周血中CD34+CXCR4+细胞显著增加(P<0.01),以S0组上升最明显,随后逐渐下降,S7组恢复至基础水平。S0、S1、S4、S7组大鼠颈总动脉样本均可见SDF-1αmRNA和蛋白表达,而C组颈总动脉样本则未见SDF-1αmRNA及其蛋白的表达。结论血管损伤后,SDF-1αmRNA和蛋白的表达显著增加,并伴随外周血中CD34+CXCR4+细胞数的增加。提示SDF-1α/CXCR4间的相互作用可能在血管损伤后的修复中发挥一定的作用。
Objective To observe the expression of stromal cell derived factor-1v (SDF-1α) in vessels after common carotid artery balloon injury. Methods Sixty male SD rats were randomly divided into control group (group C ) and balloon injury group (group S). The latter was subdivided into group SO (just after surgery), group S1 (1 d after surgery), group S4 (4 d after surgery) and group S7 (7 d after surgery). Rats in group S were performed left common carotid artery balloon injury. For all the rats, peripheral blood samples were taken, and CD34+ CXCR4+ cells were detected with flow cytometer. The left common carotid arteries were obtained to detect the expression of SDF-1α mRNA and protein by RT-PCR and Western blotting, respectively. Results The number of CD34 + CXCR4 + cells increased significantly in peripheral blood ( P 〈 0.01 ) , with that in group SO being the largest, and then fell gradually, with that in group S7 recovering to the baseline level. The expression of SDF-1 a mRNA and protein was detected in Common carotid arteries in group SO, S1, S4 and S7, while no expression of SDF-la mRNA and protein was found in group C. Conclusion The expression of SDF-1α mRNA and protein increases significantly after vascular injury, companied by the increase of CD34 + CXCR4 * cells in peripheral blood. The interaction of SDF-1α/CXCR4 may play a role in the repair of vascular injury.
