摘要
目的研究寻常型天疱疮(PV)患者外周血调节性T淋巴细胞(Treg),即CD4+CD25+Foxp3+T细胞的数量及其Foxp3mRNA的表达水平,探讨其在PV发病机制中的作用。方法收集15例PV患者(PV组)和15名健康人(对照组)外周抗凝静脉血,分离外周血单个核细胞。分别用FITC标记抗CD4单抗,APC标记抗CD25单抗,PE标记抗Foxp3单抗,流式细胞仪检测分析PV患者外周血Treg在CD4+T细胞中的百分率及CD4+T细胞在淋巴细胞中的百分率;荧光定量PCR检测外周血单个核细胞Foxp3 mRNA表达情况。结果PV组外周血Treg在CD4+T细胞中百分率为(6.99±1.86)%,CD4+T细胞在淋巴细胞中的百分率为(32.78±8.33)%,对照组分别为(7.95±1.68)%和(33.45±7.32)%,两组比较差异无统计学意义(P>0.05);荧光定量PCR检测结果显示,PV组与对照组Foxp3 mRNA相对表达量分别为8.80±5.53和8.47±5.46,两组比较无显著差异(P>0.05)。结论调节性T细胞可能不直接参与PV的发生。
Objective To investigate the number of regulatory T cells (Treg, CD4 + CD25+ Foxp3 + T cells) in peripheral blood and the expression of Foxp3 mRNA in patients with pemphigus vulgaris (PV), and explore their roles in the pathogenesis of PV. Methods Peripheral venous blood was collected from 15 patients with PV ( PV group) and 15 healthy controls (control group) , and peripheral blood mononuclear cells were isolated. Anti-CD4 mAb, anti-CD25 mAb and anti-Foxp3 mAb were marked by FITC, APC and PE, respectively, and flow cytometer was employed to measure the percent of Treg in CD4+ T cells in peripheral blood and the percent of CD4+ T cells in lymphocytes. The expression of Foxp3 mRNA in peripheral blood mononuclear cells was detected by Real-time PCR. Results The percent of Treg in CD4+ T cells in peripheral blood and the percent of CD4+ T cells in lymphocytes were (6.99 ± 1.86) % and (32.78 ± 8.33 ) %, respectively in PV group, and those in control group were (7.95 ± 1.68) % and (33.45 ± 7.32 ) %, respectively. There was no significant difference between the two groups (P 〉 0.05). It was revealed by Real-time PCR that the relative expression of Foxp3 mRNA in PV group and control group were 8.80 ± 5.53 and 8.47 ± 5.46, respectively. No significant difference was found between the two groups (P 〉0.05). Conclusion Regulatory T cells may not directly take part in the development of PV.
