摘要
以多年生黑麦草的芽尖、根尖、下胚轴、成熟种子为外植体进行了愈伤组织诱导和分化的研究,结果表明:不同划割处理的成熟种子的愈伤组织诱导率不同,不同外植体的愈伤组织诱导率也不同,芽尖、根尖愈伤组织的诱导效果较差,纵向划割的成熟种子的愈伤组织诱导率及分化能力均优于下胚轴。2,4-D浓度在7 mg/L、BA浓度为0.1 mg/L时有利于成熟种子诱导产生愈伤组织,2,4-D浓度在5mg/L、BA浓度在0.05mg/L时有利于下胚轴诱导产生愈伤组织。继代培养时间对愈伤组织的分化率有很大影响,成熟种子和下胚轴在继代培养基上继代培养60d和80d时分化率最高,分别达到76%和35%,继代天数过长不利于愈伤组织的生长。
Perennial ryegrass (Loliurn perenne) was cultured for callus induction and differentiation on MS basic medium containing different concentration of 2,4-D and BA by using different explants. Results indicated that:different explants had different induction rate and differentiation rate. Shoot tip and root tip were poor in callus induction. 7mg/L 2,4-D with 0.1 mg/L BA were propitious to callus induction from mature seeds; 5 mg/L 2,4-D with 0.05 mg/L BA were propitious to callus induction from hypocotyl. Time of callus subculture had great effect on differentiation rate.
