糖尿病大鼠海马组织中周期素依赖性激酶5表达的实验研究

摘要 目的:分析糖尿病大鼠海马组织中周期素依赖激酶5(CDK5)蛋白表达情况,探讨糖尿病影响学习与记忆的机制。方法:Y-迷宫评估大鼠学习与记忆成绩,用免疫组化法和Western blot检测海马区CDK5蛋白表达。结果:DM组大鼠第4周迷宫作业错误反应次数和全天总反应时间分别为(3.43±2.20)次、(115.42±36.64)s,对照组为(2.17±1.34)次,(90.83±20.27)s;CDK5蛋白表达相对水平为15.62±7.07,对照组为20.61±5.63,两组间差异有统计学意义(P<0.05)。结论:糖尿病大鼠CDK5蛋白表达水平下降,学习记忆能力下降,提示CDK5可能参与了学习记忆过程。 To analyze the expression level of cyclin-dependent kinase (CDK5) in hippocampal neurons, and to explore the possible mechanisms of the effect on cognitive ability caused by diabetes. Methods:At different times, the special study and memory ability were estimated by using Y-maze, the expression levels of CDK5 in hippocampus were estimated by the methods of immuno-histochemistry and Western blot. Results: At the fourth week, the error number and the total reaction time were 3,43±2.20 and 115.42±36.64 in DM group, but were 2.17±1.34 and 90.83± 20.27 in control group. The relative levels of CDK5 expression were 15.62±7.07 and 20.61± 5.63 in DM and control group, respectively. There was a significant difference between of them(P〈0.05). Conclusion: The expression of CDK5 and the ability of learning and memory were decreased in diabetic rats. It is suggested that CDK5 may play an important role in the mechanism of learning and memory.
