摘要
以受1对显性基因控制的单显性细胞核雄性不育油菜为材料,运用蛋白双向电泳技术对初花期不育花蕾和可育花蕾中蛋白质表达差异进行分析.结果表明,可育花蕾中表达的蛋白质总点数高于不育系.不育花蕾和可育花蕾中共有的蛋白点为223个,不育花蕾特有的蛋白质点数为103个,而可育花蕾中特有的蛋白质点数为160个.可育花蕾中表达的特有蛋白质分子量主要分布在60kD以下的小分子量区域,30kD尤为丰富;不育花蕾中表达的特有蛋白按分子量分布相对均匀.两者表达特有蛋白都相对集中在pI6.0~7.5区域,多为中性蛋白.
A novel genic male sterile (GMS) line in Brassica napus L. was found to be controlled by a monogenie dominant gene which we have designated as MDGMS. The differences of protein expressoin in buds between MDGMS and its allelic fertile line were detected by Two-dimensional electrophoresis (2-DE) at the same stage. The results showed that the quantity of detected protein spots in fertile buds were more than that in sterile buds. The detected protein spots both in fertile and sterile buds were 223. The spots only in fertile buds were 160. However,only in sterile buds were 103. The molecular weights of special proteins in fertile buds were mainly below 60 kD,especially around 30 kD,while the ones in sterile buds were relatively uniform. The isoelectronic points (pI) of the special protein spots in both of them were mainly in 6.0-7.5.
