摘要
建立了生物检材样品中微量毒鼠强的高效液相色谱法。生物检材样品经用乙酸乙酯提取,在优化的色谱条件下,以ZORBAX Eclipse XDB-C_8(4.6 mm×150 mm,5μm)为分离柱,乙腈-水(15+85,体积比)为流动相,在220 nm波长下进行检测;方法的线性范围为5~500μg·L^(-1),相关系数为0.999 9,方法的检出限(S/N=3)为0.5μg·kg^(-1),生物检材样品(包括血浆、胃内容物、肝、心)加标平均回收率在92.3~98.5%之间。相对标准偏差为2.67%~4.53%。
A method of HPLC determination of tetramine in biological samples was reported in this paper. Tetramine in the sample was extracted with ethyl acetate and the extract was used for HPLC analysis. Separation was carried out on a C8 column (ZORBAX Eclipse XDB-C8, 4. 6 mm ×150 mm, 5 μm) with mobile phase of acetonitrile-water (15+85, V/V). UV-detection was made at the wavelength 220 nm. Linearity of the calibration curve was ranged in 5-500 μg · L^-1 with a correlation coefficient of 0. 999 9. Detection limit (S/N=3) was found to be 0. 5 μg · kg^-1. Recovery of the method was in range of 92.3%-98.5% and RSDs of the determination were in range of 2.67%-4. 53%. The proposed method has been applied to the determination of tetramine in biological samples, including blood phasma, liver, heart and gastric contents.
