摘要
目的研究溶胶-凝胶生物活性玻璃(SGBG)/聚羟基烷酸酯(PHBV)与人牙周韧带细胞(PDLCs)的生物相容性,为牙周组织工程支架的选择提供依据。方法通过体外细胞培养实验,包括采用MTT比色分析法的浸提液实验,测定材料对人牙周韧带细胞有无毒性;及人牙周韧带细胞与SGBG/PHBV体外三维培养的直接接触法,通过扫描电镜,上清液羟脯氨酸含量的测定,检测材料对细胞生长形态及分泌功能的影响。结果SGBG/PHBV对人牙周韧带细胞无毒性,细胞在SGBG/PHBV三维支架上贴附及生长良好,实验组上清液羟脯氨酸含量测定值与对照组差异有显著性,材料不影响细胞的分泌功能,同时支架材料的三维结构更能促进细胞的分泌功能。结论SGBG/PHBV作为支架材料,有良好的生物相容性,有望应用于牙周组织缺损的组织工程修复。
[Objective ] To investigate biocompatibility between sol-gel bioglass (SGBG)/poly-3-hydroxy bu- tyrate-co-3- hydroxy valerate (PHBV) and periodontal ligament ceils (PDLCs) so as to provide a basis for the choice of scaffold material of periodontal tissue engineering. [Methods] Four different concentrations of maceration extract of SGBG/PHBV were added into the culture plants, the cytotoxicity of SGBG/PHBV was evaluated by MTT assay. And PDLCs cultured in vitro were Collected and seeded on the three-dimensional scaffolds of SGBG/PHBV, cell gowth in the scaffolds was observed by scanning of electronic microscope. And the influences of the SQBG/PHBV on cell secretory function were observed by spectrophotometry which assayed the biochemical index-hydroxyproline (Hyp). [Result] SGBG/PHBV had no cytotoxicity to PDLCs. PDLCs adhered and proliferated well on the scaffold of SGBG/PHBV. The scaffold had good influences on the cell secretory function showed by the index of Hyp. [ Conclusion] For the satisfactory biocompatibility with PDLCs, SGBG/PHBV is worth of further study as the scaffolds of periodontal tissue engineering.
