摘要
目的探讨IgA肾病(IgAN)小鼠肾脏系膜细胞凋亡及其调控基因Bax和Bcl-2表达的特点及中药益肾汤对其的影响。方法用"口服牛血清白蛋白加葡萄球菌肠毒素B尾静脉注射"法复制小鼠IgA肾病模型。实验动物分为正常组、模型组、益肾汤低浓度组及益肾汤高浓度组。HE染色光镜下观察肾小球系膜细胞增殖;原位末端标记法检测小鼠系膜细胞凋亡;免疫组化SABC法观察系膜区Bax和Bcl-2的表达。结果模型组小鼠肾脏系膜细胞凋亡显著弱于正常组(P<0.05);正常组与模型组之间Bax表达差异无显著性(P>0.05),而模型组小鼠Bcl-2的表达则明显强于正常组(P<0.05);益肾汤低浓度及高浓度组之间肾小球系膜细胞凋亡率差异无显著性(P>0.05);益肾汤低浓度及高浓度组与模型组比较,Bax表达均增强(P<0.05)、而Bcl-2表达则均减弱(P<0.05)。结论IgAN小鼠存在明显的系膜细胞凋亡不足;益肾汤可通过调控系膜区Bax和Bcl-2的表达、诱导IgAN肾脏系膜细胞凋亡,进而抑制系膜增殖。
[Objective] To study the influence of TCM Yishen Decoction on the apoptosis and its modulating genes Bax and Bcl-2 of mesangial cells of mice with IgA nephropathy. [Method] The IgAN model was built by the method of oral intake of bovine serum albumin (BSA) together with the injection of staphylococcus enterotoxin B (SEB) through caudal vein. Mice were treated with TCM Yishen Decoction. The TUNEL method was adopted to detect the mesangial cell apoptosis. The Bax and Bcl-2 proteins were detected by immunohistochemistry method. [Results] The expression of Bcl-2 in kidney tissue of mice in two TCM Yishen Decoction groups were obviously lower than that of the model group (P 〈0.05). However, the apoptosis rate of mesangial cell and Bax expression of the two TCM Yishen Decoction groups were obviously higher than that of the model group (P 〈0.05). [Conclusion] TCM Yishen Decoction may modulate the apoptosis and its modulating genes Bax and Bcl-2 of mesangial cells of mice with IgA nephropathy, thus to inhibit the mesentery proliferation.
