人参皂甙对阿尔茨海默病治疗机制的实验研究

摘要 目的研究人参皂甙对阿尔茨海默病(AD)相关β淀粉样蛋白(Aβ)诱导THP-1细胞表达诱导性一氧化氮合酶(iNOS)和一氧化氮的影响。方法用免疫印迹法测定THP-1细胞iNOS表达,用Griess法测定细胞上清中一氧化氮的浓度。结果模型组iNOS的表达明显高于对照组,与模型组比较,人参皂甙可明显减少THP-1细胞iNOS的表达和一氧化氮的产生。结论人参皂甙可以抑制THP-1细胞iNOS的表达,进一步影响一氧化氮的产生而用于AD的治疗。 Objective To study the effects of ginsenoside on expression of inducible nitric oxide synthase and concentration of NO in THP-1 induced by amyloid beta( Aβ) in Alzheimer's disease. Methods The concentration of nitric oxide was determined by Griess Reagent in supernatant of THP-1. The content of iNOS was measured by western blotting. Results The expression of iNOS of model group was significantly higher than those of control group. Compared with model group, the ginseng could reduce expression of iNOS and concentration of NO significantly in THP-1. Conclusion Ginsenoside can significantly inhibit the expression of iNOS, and reduce secretions of NO,so it may provide a novel therapy for AD.
