摘要
目的:观察佐剂关节炎(adjuvant arthritis,AA)大鼠血小板参数(血小板计数PLT、血小板压积PCT、血小板平均体积MPV、血小板分布宽度PDW)和细胞因子(IL-1β、TNF-α、IL-10)的变化及新风胶囊对其的影响。方法:将40只SD雄性大鼠随机分为正常对照组、模型对照组和新风胶囊(XFC)、甲氨喋呤(MTX)、雷公藤多苷片(TPT)3个治疗组,每组8只,用弗氏完全佐剂(Freund′s complete adjuvant,CFA)0.1ml分别向除正常对照组以外的其余大鼠右后足跖皮内注射诱发大鼠产生关节炎,观察各组大鼠血小板参数(PLT、PCT、PDW、MPV)和细胞因子(IL-1β、TNF-α、IL-10)的变化。结果:1.与正常对照组相比,模型对照组大鼠血小板参数PLT、PCT及IL-1β、TNF-α显著升高,IL-10值降低(P<0.05或P<0.01)。2.与模型对照组相比,XFC、MTX、TPT组均能升高AA大鼠IL-10,降低PLT、PCT、IL-1β和TNF-α(P<0.05或P<0.01);各治疗组相比,在升高AA大鼠IL-10方面,XFC组优于其余两治疗组(P<0.05),在降低PLT、PCT、IL-1β和TNF-α值方面无明显差异(P>0.05)。3.AA大鼠的血小板参数PLT、PCT与IL-1β、TNF-α、足跖肿胀度、关节炎指数呈正相关,与IL-10值呈负相关(P<0.01或P<0.05)。结论:AA大鼠的血小板参数PLT、PCT升高,且与IL-1β、TNF-α、IL-10、足跖肿胀度、关节炎指数具有相关性;XFC在降低AA大鼠足跖肿胀度、关节炎指数的同时,也能降低PLT、PCT,其作用机制可能与XFC能够改善病情及下调IL-1β和TNF-α,上调L-10有关。
Objective: To observe the effects of effects of Xinfeng capsule(XFC) on platelet parameters(platelet count, thrombocytocrit, mean platelet volume, platelet distribution width) and on the serum levels of cytokines(IL-1β, TNF-α, IL-10) in adjuvant arthritis(AA) rats. Methods:40 SD masculine rats were averagely divided into groups of normal control, model control, XFC treatment, methotrexate(MTX) and tripterygium wilfordii polycoride tablet(TPT) alone, with 8 in each. Except for the rats of normal group, the others were intracutaneously injected with 0.1 ml of Freund's complete adjuvant in the right hindiimb. The changes of platelet parameters (PLT, PCT, PDW, MPV) were observed by Sysmex XT-2000i automatic cytoanalyze and serum cytokines(IL-1β, TNF-α, IL-10)were analyzed with ELISA kit. Results: 1. Compared with those of normal control, platelet count, thrombocytocrit, IL-1β, TNF-α of AA rats were obviously increased and IL-10 was obviously decreased in the model groups( P 〈 0.05 or P 〈 0.01).2. Compared to that of the group of model control, IL-10 was obviously increased in the groups of XFC,MTX and TPT group, with obviously decreased PLT, PCT, IL-1β and TNF-α(P 〈 0.05 or P 〈 0.01). Compared among the treated groups, XFC treatment was better than the other two regimens because of increasing serum IL-10( P 〈 0.05 ). There was no significant differences in decreasing PLT, PCT, IL-1β and TNF-α( P 〉 0.05 ). 3. PLT and PCT of the rats were positively correlated with serum contents of IL-1β and TNF-α, as well as voix pedis' swelling and arthritis index(M). In addition, IL-10 concentration in the serum was negatively correlated with PLT and PCT( P 〈 0.05 or P 〈 0.01 ). Conclusion: Both toe's and ankle' swelling and arthritis index(M)are paralelled with changes of PLT and PCT in AA rats,which may have correlation with alterations in the amount of serum IL-1β, TNF-α or IL-10. XFC can decrease PLT and PCT.The mechanism may be related to decrease IL-1β and TNF-α, and increase IL-10 by XFC treatment.
