摘要 目的:探讨环氧合酶-2(Cyclooxygenase-2,COX-2)、凋亡抑制蛋白Survivin在HCC组织中的表达情况及两者的相关性,为HCC的基因诊断和治疗提供实验依据。方法:采用流式细胞术(FCM)及免疫组化(SP)法,定量检测肝癌、癌旁及正常肝组织中的COX-2、Survivin蛋白。结果:在HCC组织中,COX-2及Survivin蛋白阳性表达显著高于癌旁及正常对照组,两者的表达显著正相关(P<0.01)。结论:COX-2、Survivin蛋白在HCC组织中过表达是反映HCC生物学行为的有效指标,两者在HCC发生、发展中可能起协同作用。 Objective: To study the expression of COX-2 and Survivin in HCC tissues and their relationship for supplying experimental evidence for gene diagnosis and treatment of HCC.Methods: The expression of COX-2 and Survivin was detected in 30 cases of hepatoceUular carcinoma and 10 normal liver tissue by Flow cytometry (FCM) and immtmohistochemistry (SP). Results: Two experimental methods showed that the positive expression of COX-2 and Survivin in HCC was significanly higher than that in normal liver tissue and there was signitlcanly positive correlation between the expression of COX-2 and Survivin. Conclusion:The hyper-expression of COX-2 and Survivin in tissue can reflex the biological behavior of HCC and there are synergistic effect between them during the development of HCC.A possible mechanism is inhibition of turaor cell apoptosis through upregulating Survivin by COX-2,and promoting abnormal cell proliferation in development of hepatocellular carcinoma.
