摘要
利用微波辅助萃取(MAE)技术对何首乌中磷脂类化合物的萃取条件进行研究,并通过液相色谱法分离测定。通过对MAE影响因素的考察,确定萃取溶剂为氯仿-甲醇(体积比1∶2)的混合溶剂,固液比为1∶20,萃取时间15 min,温度45℃。在HPLC中,采用Nucleosil 100-5硅胶色谱柱(250 mm×4.6 mm i.d.,5μm),甲醇为流动相,流速0.8 mL/min,进样量15μL,蒸发光散射检测器。
Phospholipids in Polygonum multiflorum Thunb were extracted using microwave assisted extraction(MAE)and then analyzed by high performance liquid chromatography(HPLC) in this study.In MAE,chloroform-methanol(1 ∶2,by volume) was selected as the extraction solvent with the solid to liquid ratio of 1 ∶20,extraction time of 15 min and the temperature of 45 ℃.Phospholipids were separated on a Nuclesil 100-5 column(250 mm×4.6 mm i.d.,5 μm)with methanol as the mobile phase at a flow rate of 0.8 mL/min and detected by a evaporative light-scattering detector.The method was effective and suitable for quality control of Polygonum multiflorum Thunb and related preparations.
