摘要
目的评价结缔组织生长因子(CTGF)与原发性高血压模型大鼠(SHR)心肌纤维化的关系,以及CTGF作为抗心肌纤维化靶点的敏感性。方法将32只14周龄雄性SHR随机分为SHR对照组、卡托普利组、硝苯地平组和螺内酯组各8只,SHR对照组不给予任何药物,其他3组分别灌胃给予卡托普利100 mg.kg-1.d-1、硝苯地平30 mg.kg-1.d-1、螺内酯30 mg.kg-1.d-1,共12周。同时设8只同龄雄性SD大鼠为正常对照组。免疫组化法和RT-PCR法检测各组大鼠转化生长因子β1(TGF-β1)和CTGF表达情况,MASSON染色分析测量胶原容积分数(CVF),碱水解法测定羟脯氨酸(Hypro)含量。结果SHR对照组左室重量指数(LVI)、CVF、Hypro、CTGF、TGF-β1表达均明显高于正常对照组(均P<0.01);与SHR对照组比较,卡托普利组和螺内酯组LVI、CVF、Hypro、CTGF、TGF-β1表达显著降低(均P<0.05);CTGF、TGF-β1、CVF、Hypro和LVI呈高度正相关(P<0.01)。结论CTGF与SHR心肌纤维化关系密切,卡托普利、螺内酯能抑制其表达,CTGF可作为新的抗心肌纤维化靶点。
Objective To evaluate the correlation of CTGF and myocardial fibrosis of SHR and the possibility of CTGF as a new antlfibrotic target. Methods 32 fourteen-week male SHR were randomly assigned to captopril( 100mg·kg^-1·d-^1 ) nifedipine(30 mg·kg^-1·d-^1) ,antisterone(30 mg·kg^-1·d-^1) and SHR group equivalently, 8 male SD rats with the same age as a control group. Expression of CTGF, TGF-β1 were evaluated by immunohistochemistry and RT-PCR assay. CVF was measured by MASSON staining, and the concentration of Hypro was detected by alkaline hydrolysis method. Results The LVI,CVF,Hypro and the expression of TGF-β1 , CTGF in SHR group were significantly higher than those in SD control group (P 〈0.01 ), which were obviously decreased in the captopril and antisterone groups, compared with those in the SHR group (P 〈 0.05 ). Conclusion CTGF is closely connected with myocardial fibrosis of SHR and suppressed by captopril and antisterone. It could be regarded as a new antifibrotie target.
