摘要 目的观察α-硫辛酸抗D-半乳糖所致小鼠衰老作用。方法将60只小鼠随机分为6组各10只,分别设为空白对照组,模型组,α-硫辛酸低、中、高剂量组(给药剂量分别为50,100和200 mg.kg-1)和维生素E组(100mg.kg-1)。除空白对照组外,其他5组均每天皮下注射D-半乳糖(以0.9%氯化钠溶液配制,浓度为2.5%,注射量0.5mL),连续40 d,制作D-半乳糖致衰老模型。α-硫辛酸3个剂量组同时分别灌服以0.5%CMC配制的α-硫辛酸混悬液,维生素E组灌服维生素E植物油溶液,灌胃容积均为0.2 mL.(10 g)-1;空白对照组和模型组灌服等容积0.5%CMC溶液。连续给药40 d后,测定小鼠红细胞超氧化物歧化酶(SOD)活力,血清SOD、谷胱甘肽过氧化物酶(GSH-Px)活力及丙二醛(MDA)、一氧化氮(NO)含量,脑匀浆Na+-K+-ATP酶、单胺氧化酶(MAO)活力。结果与模型组比较,α-硫辛酸可提高红细胞SOD活力,显著升高血清GSH-Px活力,升高血清SOD活力,并降低血清MDA和NO含量;显著升高脑匀浆Na+-K+-ATP酶活力,对MAO活力有降低作用。结论α-硫辛酸对D-半乳糖所致衰老模型小鼠有较好的抗衰老作用。 Objective To investigate the antiaging effect of α-lipoic acid on aging mice induced by D-galactose. Methods 60 mice were divided into 6 groups randomly: normal group, the model group, Vit E group ( 100 mg · kg^-1 ), different doses of α-lipoic acid (200, 100 and 50 mg · kg^-1 ). The aging model was established by s. c. injecting D-galactose daily, for consecutive 40 d. Mice were i. g. three doses of α-lipoic acid, and controls for 40 d. The levels of SOD in RBC, SOD,GSH-Px,MDA,NO in blood serum and Na^+ -K^+ -ATPase,MAO in brain tissue were detected. Results Compared with model group , α-lipoic acid obviously improved SOD in RBC ; content of GSH-Px, decreased MDA and NO in blood serum, and significantly stimulated Na^+ -K^+ -ATPase, suppressed MAO in brain tissue. Conclusion α-Lipoic acid shows a good antiaging action on the aging murine model induced by D-galactose.
