摘要
目的探讨抗体催化水的氧化作用对泡沫细胞形成的影响。方法将THP-1单核细胞与血浆低密度脂蛋白(LDL)、佛波酯(PMA)、人IgG共同培养,并设空白组和对照组。通过硫代巴比妥酸(TBA)法检测细胞内脂质过氧化产物,以及油红O染色检测泡沫细胞的形成,观察分析IgG对THP-1单核细胞泡沫化形成的影响。结果IgG显著促进了PMA激活的THP-1单核细胞内脂质过氧化产物的积聚和泡沫细胞的形成。结论抗体催化水的氧化作用可能是动脉粥样硬化发病过程的一种重要机制。
Objective To investigate the effects of antibody-catalyzed water oxidation on formation of foam cells.Methods THP-1 monocytes were cultured with low density lipoprotein(LDL),phorbol myristate acetate(PMA) and human IgG.Lipid peroxide determination in cells was done by thibabituric acid(TBA) method,and formation of foam cells was analyzed by the oil red O staining cells.Results The human IgG significantly promoted the accumulation of lipid peroxide in PMA-activated THP-1 monocytes and the formation of foam cells.Conclusion antibody-catalyzed water oxidation could be a new mechanism involved in atherosclerosis.
