摘要
以菊粉为碳源,研究了产菊粉酶酵母HKD-06的最佳细胞培养条件。结果表明:适宜的菊粉浓度为100 g/L;最适氮源为酵母粉5 g/L+蛋白胨2 g/L;最适生长温度30℃;最适生长pH8.0。研究发现高浓度(150 g/L)菊粉不会造成底物抑制现象,同时也不会造成搅拌及溶氧的困难;培养基中的金属离子对该酵母细胞生长有显著的影响,Fe2+可促进酵母细胞的生长,而Mg2+、Zn2+、Cu2+却表现出显著的抑制作用。
The optical cell cultivation conditions of yeast HKD-06 which produced inulinase with iunlin as carbon sources were studied. The results showed that the optical inulin concentration was 100 g/L, the optical nitrogen sources were yeast extract 5 g/L and peptone 2 g/L, optical cultivation conditions were 30 ℃ and pH 8.0. It was found that high concentration of inulin (150 g/L) caused neither substrate inhibition on cell growth nor difficuhies of agitation and oxygen dissolution. It was also found that metal ions in the media obviously affected the cell growth of yeast, Fe^2+ could stimulate the yeast cell growth, and Mg^2+, Zn^2+, Cu^2+ had great inhibition effects.
