摘要
研究了卡那霉素和羧苄青霉素对油葵不定芽的诱导及生长的影响。在以卡那霉素为抗性选择标记时,对油葵不定芽起完全抑制作用的Km临界浓度在75~100mg/L;在以羧苄青霉素为抑菌剂用于油葵遗传转化试验时,其浓度为300mg/L时即可有效抑制农杆菌的生长,同时又不会对不定芽的诱导及生长产生明显的伤害作用。试验结果表明,卡那霉素及羧苄青霉素均可作为油葵转化植株的有效筛选、抑菌剂。
In the exeperiment,the optimal concentrations of kanamycin and carbenicillin in the selected medium were investigated. The results indicated that the inhibited concentrations of kanamycin of adventitious bud formation of Helianthus annus were 75 mg/L and 100 mg/L,when the carbenicillin was used in the transformation procedure;the concentrations was 300 mg/L when the growth of agrobacterium could be effectively inhibited. At the same time,the induction and growth of adventitious bud were not remarkably injured. The experiment results indicated that both kanamycin and carbenicillin could be selected as bacteriostatic for transforming Helianthus annus into plant.
