支气管哮喘小鼠肺组织及肺T细胞中Notch3及Foxp3的表达及意义 被引量:7

Expressions of Notch3 and Foxp3 in Lung Tissues and T Cells Isolated from Lungs in Asthmatic Mouse Models and Its Significance

摘要 【目的】研究支气管哮喘模型小鼠肺、肺T细胞Notch3 mRNA及CD4+CD25+调节性T细胞的特征性标志物Foxp3 mRNA的表达特征,探讨Notch3与CD4+CD25+调节性T细胞的关系及其在支气管哮喘发病中的作用。【方法】制作哮喘小鼠模型;分离肺中T细胞;利用RT-PCR半定量技术,与对照组比较,测定小鼠肺组织、肺T细胞Notch3 mRNA及Foxp3 mRNA的表达。【结果】哮喘组及对照组小鼠肺组织及肺T细胞中Notch3 mRNA及Foxp3 mRNA均有表达,哮喘组小鼠肺组织及肺T细胞中Notch3 mRNA及Foxp3 mRNA的表达较对照组均有降低。【结论】Notch3信号对CD4+CD25+调节性T细胞有一定的调节作用,并且在支气管哮喘的发病机制中可能发挥一定作用。 [Objective]To investigate the expressions of Notch3 and Foxp3 considered as a specific marker for CD4^+ CD25^+ regulatory T (Treg) cells in lung tissues and T cells isolated from lungs in asthmatic mouse models and the relation between Notch3 and CD4^+ CD25^+ Treg cells,as well as the role of them in the pathogenesis of asthma. [Methods]Asthmatic mouse models were made. T cells were isolated from lungs and their purity was identified. The mRNA expressions of Notch3 and Foxp3 in lung tissues and T cells isolated from lungs were detected by reverse transcription polymerase chain reaction (RT-PCR). [ResultslThe expressions of Notch3 and Foxp3 could be detected both in lung tissues and T cells isolated from lungs, and they both had a significantly decreased expression in asthmatic group compared with control group.
