摘要
目的研究高胆固醇血清对血管内皮细胞VEGF表达的影响以及辛伐他汀的抑制作用。方法分别培养的血管内皮细胞予不同浓度的高胆固醇血清或合用100nmol/L的辛伐他汀共同培养,分别测定其培养上清液中VEGF水平。结果当胆固醇浓度大于0.08mmol/L时,血管内皮细胞VEGF表达显著增加;且随胆固醇浓度的增加,VEGF的表达亦显著增强。但可被100nmol/L的辛伐他汀所抑制,不同高胆固醇血清合用辛伐他汀组其血管内皮细胞VEGF表达与正常对照组无差异。结论高胆固醇血清可致血管内皮细胞VEGF表达增高,而高胆固醇的上述作用可被辛伐他汀显著抑制。
Objective To observe the effect of hypercholesterolemia on the expression of VEGF in endothelial cells and the role of simvastatin. Methods Cultured endothelial cells were treated with different concentration of hypercholesterolemic rabbit serum or combined with 100mnol/L simvastafin. Culture supernatant was collected for VEGF detection by ELISA. Results VEGF expression in endothelial cells treated with hypercholesterolemic serum (cholesterol concentration 〉 0.08 mmol/L) was significantly higher than that treated with control. 100nmol/L Simvastatin could significantly suppress the enhanced VEGF expression induced by hypercholesterolemic rabbit sertan. Conclusions Hypercholesterolemic serum could significantly enhance VEGF expression in endothelial cells, which could be suppressed remarkably by simvastatin,
