摘要
目的:观察小鼠围植入期子宫内膜组织中白血病抑制因子(LIF)表达的变化规律并探讨其与胚胎种植的关系。方法:取真孕、假孕第3、4、5、6天小鼠(n=10)子宫内膜组织,采用免疫组织化学、RT-PCR法分别检测LIF蛋白和mRNA的表达情况,并设未孕小鼠作对照组。结果:小鼠子宫内膜组织中LIF蛋白的表达主要定位在腔上皮和腺上皮。真孕组第3、4、5、6天子宫内膜组织中LIF蛋白和mRNA的表达水平均高于假孕组和对照组(P均<0.05),且表达水平在孕4d达高峰(P<0.05)。结论:LIF表达的峰值与胚胎种植时间一致,可能是子宫内膜容受性的标志;LIF的表达不仅受母体激素的影响,也和胚胎的刺激有关。
Aim: To evaluate the expression regulation of LIF in peri-implantation mouse endometrium tissue and ex- plore the relationship between LIF and blastocyst implantation. Mothods:Endometrium samples were collected from pregnant and pseudo-pregnant mice on day 3,4,5 ,and 6 of gestation. LIF protein and its location were determined by immunohistochemistry. The LIF mRNA expression was measured by RT-PCR. Results: Immunostaining of LIF protein was located in endometrial epithelial cell and glandular epithelial cell. In the pregnant group, the levels of LIF protein and mRNA on day 3 - 6 were significantly higher than that in pseudo pregnant group and non-pregnant group (P 〈 O. 05 ). Both LIF protein and mRNA levels peaked on day 4 of gestation ( P 〈 O. 05 ). Conclusion : LIF takes part in the process of blastocyst implantation,and may be the candidate markers of uterine receptivity. The expression of LIF is regulated not only by maternal hormone, but also by the implanted embryos.
