摘要
目的:应用高效液相色谱-电喷雾离子阱质谱法鉴定复方通关丸中的化学成分。方法:实验采用DiamonsilTMC18色谱柱,实验条件Ⅰ:水(A)/乙腈(B)梯度色谱分离,负离子模式下采集数据;实验条件Ⅱ:0.3%冰醋酸水溶液(A)/甲醇(B)梯度色谱分离,正离子模式下采集数据。结果:根据正、负离子模式下的准分子离子峰和二级质谱,通过与文献数据或部分标准品对照,鉴定了12个化合物,分别是新芒果苷、芒果苷、知母皂苷B-Ⅱ、知母皂苷E、知母皂苷B-Ⅲ、知母皂苷A-Ⅲ、黄柏碱、木兰花碱、蝙蝠葛任碱、小檗碱、药根碱和巴马汀。结论:HPLC-MS/MS为中药复方中化学成分的鉴定提供了一种快速准确的方法。
AIM： To identify the chemical components of Tongguanwan prescription by high performance liquid chromatography-electrospray ion trap-mass spectrometry （HPLC-ESI/IT-MS）. METHOD： The chromatographic conditions were as follows：Diamonsil^TM C 18 （150mm×4.6 mm, I.D.5μm） column; mobile phaseⅠ ： water （A） and acetonitrile（B）,gradient elute,the ESI source was used and the data in negative mode were acquired; mobile phase Ⅱ： water containing 0.3% acetic acid（A） and methanol（B）,gradient elute, the data in positive mode were acquired. RESULT： The quasi-molecular ions of compounds in both negative and positive modes were observed for molecule mass information, and the potential structures were elucidated by analyzing the mass spectra of compounds through comparing them with the reference data and some standards. Twelve compounds were identified in the prescription, which were neomangiferin, mangiferin, timosaponin B-Ⅱ, anemarsaponin E, timosaponin B-Ⅲ, timosaponin A-Ⅲ, phellodendrine, magnoflorine, menisperine, berberine, jatrorrhizine and palmatine. CONCLUSION： HPLC-MS/MS is a rapid and accurate method to identify the compositions of prescription of traditional Chinese medicine.
