HPLC法测定乳癖安消颗粒中盐酸小檗碱的含量

Determination of berberine hydrochlodride in Rupi Anxiao Granules by HPLC

目的建立高效液相色谱法测定乳癖安消颗粒中盐酸小檗碱含量的方法.方法采用高效液相色谱法,以十八烷基硅烷键合硅胶为填充剂,流动相采用乙腈-0.05mol·L-1磷酸二氢钠溶液(用磷酸调节pH值至3.0)(30∶70);流速:1mL·min-1;检测波长:265nm;柱温:35℃.结果在此色谱条件下,线性范围为10～70μg·mL-1,r=0.9999,样品平均回收率为99.87%.结论本方法简便、灵敏度高、分离度好,可控制乳癖安消颗粒中盐酸小檗碱的含量.

OBJECTIVE To establish an HPLC method for determination of berberine hydrochlodride in Rupi Anxiao Granules. METHODS Kromosil C18 column（4.6mm × 150mm,5um） was used, acetonitrie--0.05mol · L^-1 phosphate buffer of pH 3.0（30 ： 70） as mobile phase and the UV detection wavelength was 265nm,the flow rate was 1mL · min^-1 and the column temperature was 35℃. RESULTS Under the chromatograph conditions,there was a good linear relationship between 10-70ug · mL^-1 , r =0. 9999, the average recovery was 99.87% . CONCLUSION This method was available,simple and accurate,and can be used to control berberine hydrochlodride contents in Rupi Anxiao Granules.
