摘要
目的寻找一种胰腺冰冻切片免疫组化过程中的最佳组织固定方式。方法将20例冰冻组织OCT包埋后置于冰冻机切片8~10μm,每例组织各切5张片,各分成5组,接着分别采用以下5种不同的方法进行固定10min,即丙酮4°C固定法(A组)、丙酮室温固定法(B组)、4%多聚甲醛室温固定法(C组)、10%甲醛室温固定法(D组)、Bouin液室温固定法(E组),然后0.01MPBS冲洗5min×3次,枸橼酸电炉热修复后进行免疫组化染色。结果A组的冰冻切片免疫组化染色结果比其他组好。结论丙酮4°C固定比4%多聚甲醛、10%甲醛、Bouin液固定较好保存胰腺中PKCa抗原的活性。
Objective To find a frozen section immunohistochemistry pancreas in the process of fixation of the best organizations. Methods 20 cases of frozen tissue was embed by OCT before placing frozen-embedded sections 8-10μm, cut each of the five organizations film, divided into five groups, and then each of the following five different methods of fixation, namely, acetone 4°C fixation (Group A), acetone fixed at room temperature (Group B), 4% more than POM fixed at room temperature (Group C), 10% formaldehyde fixed at room temperature (Group D), fixed Bouin liquid at room temperature ( Group E), 0.01M PBS and then washed 5minx3 times, heat repair by furnace after immunohistochemical staining. Results Group A frozen section immunohistochemical staining results was better than any other group. Conclusion 4°C acetone fixed was better than 4% POM, 10% formaldehyde, better preservation of Bouin PKCa antigen activity.
