乳腺癌转移抑制因子1在乳腺癌中的表达及其与雌激素孕激素受体的相关性 被引量:4

摘要 目的探讨乳腺癌转移抑制因子(BRMS1)在乳腺癌中的表达及其与雌激素受体(ER)、孕激素受体(PR)的相关性。方法提取组织总RNA,将mRNA转录成cDNA,然后用实时荧光定量聚合酶链反应(RFQ-PCR)检测乳腺组织BRMS1 mRNA表达,以BRMS1 mRNA和3-磷酸甘油醛脱氢酶(GAPDH)mRNA含量的比值表示BRMS1表达水平。免疫组化检测ER、PR。结果乳腺癌组织中BRMS1表达水平明显低于癌旁组织(P<0.01),也明显低于正常乳腺组织(P<0.01),BRMS1低表达与临床病理分期和淋巴结转移有关,BRMS1在ER阳性乳腺癌组织中的表达高于在ER阴性组织中的表达(P<0.05),BRMS1在PR阳性乳腺癌组织中的表达与在PR阴性组织中的表达比较,差异无统计学意义(P>0.05)。结论乳腺癌组织中BRMS1的低表达与乳腺癌的发展与转移有关,BRMS1表达水平与ER有一定相关性。 Objective To study breast cancer metastasis suppressor 1 (BRMS1) gene expression in breast cancer and its relationship with estrogen receptor (ER) and progesterone receptor (PR). Methods Total RNA was extracted and mRNA was transcribed reversely into cDNA, and then real-time fluorenscence quantitative polymerase chain reaction (RFQ-PCR) was used to detect BRMS1 mRNA in breast tissue. BRMS1 gene expression was presented as the ratio of BRMS1 mRNA to GAPDH mRNA. ER and PR were detected with immunohistochemical assay. Results The expression level of BRMS1 was significantly lower in breast cancer tissue than that of the adjacent tissue (P〈0.01) and normal lung tissue (P〈0. 01). BRMS1 low expression was related to pathologic staging and lymph node metastasis. BRMS1 expression level in ER (+) cancer tissue was significantly higher than that of ER (-) cancer tissue (P〈0.05) ; while the difference in its expression level wasn't statistically significant between PR (+) and PR(-) cancer tissue (P〉0.05). Conclusion BRMS1 low expression may play a crucial role in the progression of breast cancer and lymph node metastasis,and BRMS1 expression leyel is asoociated with ER.
