Cx26在神经管周围间充质细胞的表达及意义

摘要 目的观察Cx26在神经管畸形发生中神经管周围间充质的表达,以探讨Cx26与NTDs发生的关系。方法用过量RA建立小鼠胚胎NTDs模型。用SABC免疫组化方法和图像处理系统,分析比较实验组和对照组神经管发育情况以及Cx26蛋白表达的位置和强度。结果Cx26蛋白表达于神经上皮细胞的细胞膜和细胞质内,而且实验组显著高于对照组(P<0.05);Cx26蛋白从E8 d到E11 d表达逐渐增多,E11 d达到高峰,然后逐渐下降。结论Cx26蛋白定位于神经管上皮细胞的胞膜和胞质内,其表达有一定的规律性。Cx26蛋白的过高表达可能与RA致NTDs密切相关。 Objective To explore the expression of connexin 26(Cx26) and its role in the pathogenesis of neural tube defects(NTDs). Methods NTD Models in mouse embryo were set up by overdose of ratinoic acid (RA) to analyze and compare neural role developmental conditions, the position of expression and the intensity of Cx26in both experimental and contrast groups by means of SABC immunohistochemical method and image processing system. Results The expressions of Cx26 were not only located in membranes, but also in the cytoplasms of neural epithelia. The positive rate of Cx26 expressions was higher in the experimental group ( P 〈 0.05). The expressions of Cx26 gradually increased from E8 d to Elld, reached the peak value at Elld and then declined. Conclusion The Cx26 positive immunostaining is located in both membranes and the cytoplasms of neural epithelia. The expressions seem regular to some extent. The results indicate that the hypsi-expressions of Cx26 may play an important role in the development of TNDs induced by RA.
