摘要
目的考察姜黄素对原代培养的肾母细胞瘤细胞的影响及其相关机制。方法用MTT法和Giemsa染色观察姜黄素对原代培养的肾母细胞瘤细胞增殖的抑制作用,DNA片段化实验观察姜黄素的诱导凋亡作用,蛋白印迹和免疫组化法观察姜黄素对肾母细胞瘤细胞Bcl-2和Caspase-3表达的影响。结果姜黄素对原代培养的肾母细胞瘤细胞有明显的增殖抑制作用,呈剂量依赖性。姜黄素浓度为3.68μg/ml和7.36μg/ml时均可诱导细胞凋亡。与阴性对照组相比,3.68μg/ml和7.36μg/ml的姜黄素对Bcl-2表达的影响不明显,但使Caspas-3表达明显升高。结论姜黄素可诱导肾母细胞瘤细胞凋亡,姜黄素对肾母细胞瘤的促凋亡作用部分是通过对Caspas-3的调控来实现的,Bcl-2的表达可能对肾母细胞瘤的凋亡不起主要的作用。
Objective To investigate the effect of curcumin on primarily cultured Wilm's tumor ceils. Methods Wilm's tumor cells were primarily cultured with different concentrations of curcumin. MTT and Giemsa staining were used to examine cell proliferation. Cell apoptosis was detected by DNA fragmentation method and the expression of Bcl-2 and Caspase-3 was measured by Western blot and immunohistochemistry. Results Curcumin inhibited Wilm's tumor cells in a dose-dependent manner. Curcumin at 3.68 μg/ml and 7.36 μg/ml induced Wilm's tumor cells apoptosis as shown in DNA ladder display. Bcl-2 had no changes after treated with curcumin at 3.68 μg/ml and 7.36 μg/ml for 24h, but the expression of Caspase-3 increased. Conclusion Results suggest that curcumin can induce apoptosis of Wilm's tumor cells, which is associated with the expression of Caspase-3, but not expression of Bcl-2.
