2种血小板低渗休克反应测定方法的比较研究 被引量:8

摘要 目的建立生化分析仪测定血小板低渗休克反应(HSR)的方法,并与血小板聚集仪法进行比较。方法分析参数设置:试验模式为实时吸光度监测,测定温度37℃,测定波长610nm,样品与试剂比例2∶1,延迟时间5s,监测时间15min。制备含不同比例球形血小板的富含血小板血浆(PRP),对分析方法进行敏感性、准确性、重复性评价。结果生化分析仪法和血小板聚集仪法检测HSR的敏感度均为20%;对含高、中、低比例球形血小板样品重复5次的变异系数(CV)分别为9.4%、4.6%、3.8%和11.0%、4.8%、4.2%;HSR测定值与球形血小板的含量密切相关,其相关系数(r)分别为0.9668和0.9504。结论应用生化分析仪测定HSR,其检测敏感性与血小板聚集仪法相同,重复性、准确性均可达到方法学要求,可用于HSR的自动化检测。
