摘要
目的:探讨低能量激光照射联合复合多糖对荷肝癌小鼠肿瘤细胞周期及凋亡的影响。方法:建立荷肝癌小鼠皮下种植瘤模型,将荷瘤小鼠随机分为5组,分别为NS组、低能量激光照射组、复合多糖组、低能量激光照射联合复合多糖组及5-氟尿嘧啶(5-fluorouracil,5-FU)组。对照组给生理盐水灌胃,治疗组给复合多糖、5-FU灌胃,用低能量激光照射脾区进行治疗,连续治疗10d后将小鼠处死。取肿瘤组织用流式细胞仪检测细胞周期及细胞凋亡;免疫组化法检测瘤组织中凋亡抑制因子survivin蛋白的表达情况。结果:低能量激光照射联合复合多糖可以使肿瘤细胞停止于G1期,促进肿瘤细胞凋亡,并显著下调survivin蛋白的表达。结论:低能量激光照射联合复合多糖能够调控肿瘤细胞周期、诱导肿瘤细胞凋亡,抑制荷肝癌小鼠肿瘤的生长。
Objective:To study effects of low energy laser radiation unite complex polysaccarides on tumor cell cycle and cell apoptosis in liver cancer loaded mouse. Methods:Liver cancer loaded mice models were established and divided randomly into five groups, namely, NS group, low energy laser radiation group, complex polysaccharide group, low energy laser radiation unite complex polysaccharide group and 5 - FU (5 - fluorouracil, 5 - FU ) group. NS was given to the control group orally, complex polysaccharide and 5 - FU were given to the experimental groups. The spleen was lighten with the low energy laser radiation every day. Then tumors were taken out after 10 day treatment. Cell cycle and apoptosis were detected with flow cytometry. The expressions of survivin protein were detected with immunohistochemistry. Results: low energy laser radiation unite complex polysaccharide could arrest tumor cells at G1 stage, induce tumor cells apoptosis, and decrease the expression of survivin protein. Conclusion:low energy laser radiation unite complex polysaccharide can regulate the tumor cell cycle, induce tumor cell apoptosis and inhibit the growth of the liver cancer in mice.
