摘要
目的观察碱性成纤维细胞生长因子(bFGF)转染自体骨骼肌干细胞缺血心肌内移植对心肌结构及骨骼肌卫星细胞转归的影响。方法取大白兔大腿肌,分离卫星细胞后培养、转染并用4',6-二乙酰基-2-苯基吲哚(DAPI)标记。建立急性心肌梗死动物模型。将bFGF转染的卫星细胞注射在梗死灶周边心外膜下。4周后取出心脏,对缺血心肌结构及卫星细胞转归情况进行观察。结果移植的卫星细胞分化成为带有横纹的心肌样细胞;在卫星细胞移植区心肌细胞玻璃样变性较轻,肌纤维排列有序,微血管生长良好。结论bFGF转染自体骨骼肌卫星细胞在缺血心肌中可分化成心肌细胞样细胞,局部血液循环得到改善。
Objective To observe the influence of transfected autologous skeletal muscle stem cell with bFGF to rabbit ischemic myocardial structure. Methods Satellite cells isolated from rabbit skeletal muscle, cultured, transfected with bFGF and labeled by 4' ,6-diamidino-2- phenylindole (DAPI), then iniected into ischemic myocardium surrounding. The specimens were harvested 4 weeks letter. Histologic sections were examined under common light microscope and fluorescent microscope. Results The progressive differentiation of implanted satellite cells into fully developed striated muscle fibers could be observed. The fibroatrophy of the host ischemic myocardium had been effectively inhibited, muscle fibers arranged in an orderly manner and microvascular growth well. Conclusion Transfected autologous skeletal muscle stem cell with bFGF implanted into the ischemic myocardium of rabbit can differentiate from cardiomyocytelike cells. The local blood circulation is improved.
