摘要
实验将DAPI染色方法运用到小鼠微核检测中。选取20只小鼠随机分为实验组和对照组,每组10只。2组均往小鼠腹腔注射环磷酰胺60mg/kg,1次/d,连续注射3d。实验组采用DAPI染色;对照组采用10%的Giemsa染色。结果显示,实验小鼠骨髓组织细胞微核DAPI染色特异性强,易判断,对照组Giemsa染色结果特异性差,且不能排除染色过程中Giemsa染液中的染色颗粒沉淀的干扰,这样就使Giemsa染色后,对微核的判断造成困难。本实验建立了小鼠骨髓组织细胞微核DAPI染色方法,检测效果好,特异性强。
The DAPI sta/n method was applied in mouse micronucleus examination. A total of 20 mice were equally and randomly divided into an experimental group and a control group. 60 mg/kg cyclophosphamide was intraperitoneally injected into mice, once a day for successively 3 days in both groups. DAPI stain was applied in the experimental group. Giemsa (10%) stain was applied in the control group. Results showed that the specificity of the micronucleus on bone marrow cells with DAPI stain was stronger and could be easily judged. While the specificity of the Giemsa stain in the control group was poor. The interference of the staining particle in Giemsa staining solution was not been removed during the staining process. Thus, the judgment of the micronucleus is difficult after the Giemsa stain. This study established the method of micronucleus with DAPI stain with mouse myeloid tissue cells, resulting in a good effect and strong specificity.
