摘要
目的了解引导神经发育的Robo4基因在实体肿瘤血管新生中的表达,证实Robo4是肿瘤血管新生特异性标志物(tumor endothelial marker,TEM)。方法采用RT-PCR技术检测多种不同细胞中Robo4 mRNA的表达情况;采用原位杂交和免疫组化染色检测多种肿瘤组织及其对应正常组织的相邻组织切片中Robo4和CD31在微血管的表达情况。结果RT-PCR显示在包括肿瘤细胞和正常细胞的9种受检细胞株中,Robo4 mRNA特异性表达于血管内皮细胞HUVEC和HMVEC中;Robo4在各种正常组织的微血管中表达极其微弱,而在肿瘤组织的微血管上显著表达;CD31在正常和肿瘤组织的微血管上均显著表达。结论Robo4仅特异性表达于肿瘤血管新生的部位,是肿瘤血管新生标志物。
Objective To investigate the expression patter of Robo4 during tumor which guides neurons to an appropriate destination during CNS development. Methods anglogenesls, RT-PCR was used to examine Robo4 mRNA expression in different cell lines. In situ hybridization and immunohistochemistry staining were employed in different tumor and accordant normal tissue samples to detect the expression of Robo4 and CD31 in adherent sections. Results In nine checked cell lines, Robo4 mRNA specifically expressed in HUVEC and HMVEC, which are of vessel endothelial origin. In situ hybridization and immunohistochemistry staining showed obvious expression of Robo4 in tumor angiogenic sites while the expression of Robo4 in normal tissues was very faint. CD31 expression were apparent both in tumor and normal tissues. Conclusions Robo4 is specifically expressed in tumor angiogenic sites and it is a tumor endothelial marker.
