摘要
背景与目的:卡铂作为第二代铂类抗肿瘤药,已越来越多地应用于临床多种恶性肿瘤的治疗。但大剂量应用时骨髓抑制明显,结合放疗时更为显著。本文通过动物实验,探讨卡铂分次给药联合X线治疗小鼠Lewis肺癌时对其免疫功能的影响。方法:采用在小鼠右前腋窝接种Lewis肺癌细胞的荷瘤小鼠模型,观察在总剂量相同情况下,卡铂单次与分次给药联合X线照射小鼠Lewis肺癌时对肿瘤生长及其免疫功能的影响。结果:对照组小鼠血清IL-10、TNF-α、脾指数分别为(144.9±48.4)ng/ml、(194.63±64.4)ng/ml、(12.07±2.2)mg/g;卡铂单次给药+X线组分别为(279.0±46.9)ng/ml、(71.5±8.4)ng/ml、(5.52±1.31)mg/g,与对照组相比,IL-10水平明显升高、TNF-α水平和脾指数降低(P<0.001);卡铂分次给药+X线组分别为(209.0±60.6)ng/ml、(121.9±32.7)ng/ml、(7.67±1.23)mg/g,与对照组相比,亦是IL-10水平明显升高、TNF-α水平和脾指数降低(P<0.05),该对比结果相对于卡铂单次给药+X线组与对照组比较的结果,对小鼠IL-10的含量、TNF-α的含量和脾指数影响则明显减轻,差异有显著性(P<0.05)。结论:在总剂量相同情况下,卡铂分次给药结合X线照射,较单次给药对小鼠机体的免疫功能抑制作用要小。
Background and purpose: As the second general platinum antineoplastic agent, carboplatin has been applied in treating more and more malignant tumors clinically. But it has severe bone marrow suppression when applied in large doses. The purpose of this research was to explore the influence of carboplatin of fractional dose, combined with X ray, on the immunity of Lewis lung cancer in mice. Methods: The model of tumor-bearing mice was induced by injecting Lewis lung cancer cells into the right infra-axillary dermis. In cases given the same total dose, the tumor growth and the immune suppression effect of carboplatin of single dose or of fractional dose, combined with X ray radiation, on the mouse with Lewis lung cancer were observed. Results: The concentration of IL-10 and TNF-α in serum,the spleen exponent were respectively( 144.9±48.4)ng/ml,( 194.63±64.4)ng/ml,( 12.07±2.2)mg/ g in the control group;(279.0±46.9)ng/ml,(71.5±8.4)ng/ml,(5.52±1.31)mg/g in carboplatin of single dose with X ray radiation group, which ,compared with the control group, can dramatically increase the concentration of IL-10,decrease the concentration of TNF-α in the serum ,and dramatically decrease the spleen exponent(P〈0.001);(209.0±60.6) n g/ml,(121.9±32.7)ng/ml,(7.67±1.23)mg/g in the carboplatin of fractional dose with X ray radiation group, which, compared with the control group, can also dramatically increase the concentration of IL-10 ,decrease the concentration of TNF-α in the serum ,and dramatically decrease the spleen exponent(P〈0.05).But, compared with the carboplatin of single dose with X ray radiation group ,it had a lighter influence on the concentration of IL-10,TNF-α in the serum and the spleen exponent(P〈0.05),the difference was of statistical significance. Conclusions: In cases given the same total dose, compared with the single dose, the suppression effect of carboplatin of fractional dose ,combined with X ray radiation, on the immune function of mouse was lighter.
