补肾益气和血方对胚泡着床障碍小鼠子宫内膜腺体凋亡的影响

Effect of Bushen Yiqi Hexue Recipe on the Endometrial Gland Apoptosis in Blastocyst Implantation Dysfunctional Mice

目的观察补肾益气和血方对胚泡着床障碍小鼠子宫内膜腺体凋亡的影响。方法复制小鼠胚泡着床障碍模型,分为对照组、模型组和治疗组,每组30只。观察小鼠妊娠率和着床胚泡数,分析子宫内膜腺体的凋亡情况,检测子宫内膜腺体的增殖细胞核抗原(PCNA)、bax、bcl-2和激活型Caspase-3蛋白表达情况。结果模型组妊娠率和平均着床胚泡数均显著降低,治疗组两者均较模型组显著提高(P<0.05或P<0.01)。模型组子宫内膜腺体增殖指数(PI)明显高于对照组(P<0.01),而凋亡指数(AI)则明显低于对照组(P<0.05);治疗组与模型组比较,腺体PI明显降低而AI明显增高(P<0.01),两者与对照组比较差异均无统计学意义。模型组子宫内膜腺体中bax和激活型Caspase-3蛋白显著低于对照组(P<0.01);治疗组两者则显著高于模型组(P<0.01);治疗组与对照组差异无统计学意义。结论促进子宫内膜腺体中bax蛋白的表达和Caspase-3的激活,调节植入窗期腺体细胞增殖和凋亡的平衡,可能是补肾益气和血方促进子宫内膜接受性建立和改善胚泡着床障碍的机制之一。

Objective To observe the effect of Bushen Yiqi Hexue Recipe （Recipe to reinforce kidney, replenish qi and harmonize blood） on the endometrial gland apoptosis in blastocyst implantation dysfunctional mice. Methods The mice models of blastocyst implantation dysfunction were made and divided into control, model and treated groups with 30 in each. Pregnancy rate, number of implantation blastocysts, the expression of proliferating cell nuclear antigen （PCNA）, bax, bcl-2 and activated Caspase-3 were observed. Results Compared with the control group, pregnancy rate and mean implanted blastocysts in the model group were remarkably reduced. Compared with the model group, pregnancy rate and mean implanted blastocysts in the treated group were significantly improved （P〈0.05, P〈0.01）. Comparing with the control group, proliferation index （PI） of endometrial gland in the model group was dramatically elevated （P〈0.01）, whereas apoptosis index （AI） was dramatically reduced （P〈0.05）. Comparing with the model group, PI of endometrial gland in the treated group was obviously inhibited, while AI was significantly improved （P〈0.01）, both showed no remarkable difference from those of control group. Bax and activated Caspase-3 protein expressions of endometrial gland in the model group was remarkably lower than in the control group （P〈 0.01）, and these expressions in the treated group was significantly higher than in the model group （P〈0.01）, but there was no significant difference in comparison of the treated group with the control group. Conclusion It is suggested that increasing bax and activated Caspase-3 expression and regulating the imbalance of apoptosis and proliferation in endometrial gland may be one of the mechanisms of Bushen Yiqi Hexue Recipe in improving uterine receptivity and eventually promoting blastocyst implantation.
