摘要
对来自武汉市郊某养鸽专业户及市场食用鸽共70只采集口腔咽部粘膜拭子及其中15只鸽子的气管、气囊和肺等材料进行了支原体的分离鉴定,其中口腔咽部拭子几乎100%能分离到支原体,但均与细菌共存而不易纯化,其他部位均未分离到支原体。对已纯化的34个分离物中的19个进行了鉴定,用鸽支原体MMP1株、鸽口支原体MMP4株及鸽鼻支原体694株作为标准抗原制备抗血清,以代谢抑制试验进行了血清学鉴定。结果表明,所有分离物均能在修改FM-4培养基上生长,在固体培养基表面形成典型的“油煎蛋”状或“脐状”菌落;光镜下和电镜负染后观察,菌体多呈现球状、点状等多形态,经超薄切片后观察,菌体外无细胞壁包绕,仅有由3层单位膜构成的细胞膜结构,所有分离物在无醋酸铊及青霉素的培养基上连续传5代不回复为细菌形态,能在含有猪血清的培养基上生长,其中5株水解精氨酸而不发酵葡萄糖,14株发酵葡萄糖但不水解精氨酸。经抗血清代谢抑制试验鉴定,前者为鸽支原体,后者为鸽口支原体。
Swabs of oropharynxes,tracheas,air sacs and lungs from seventy pigeons in Wuhan were subjected to isolation of mycoplasmas.Nineteen isolates were identified through biochemical,serological and electron microscopic methods recommended by Subcommittee on the Taxonomy of Mollecuttes,International Commettee on Systematic Bacteriology.All pured isolates could form typical“fried egg”or nipple colonies on agar media and showed very small size and pleomorphic shape under low amplificattion microscope.Electron microscopy studies revealed that pleomorphic cells were surrounded by a triple layer membrane.The microorganism did not revert to bacterial form through five consecutive subcultures in media free from antibiotics.these findings reveal that nineteen isolates belong to Class Mollecutes,Order Mycoplasmatsle,Family Mycoplasmataceae.By the metabolish inhibitation test,five of the nineteen isolates are proved to be Mycoplasma columbinum and the others to be Mycoplasma columborale.
