软肝冲剂对肝硬化患者外周血单核细胞TGF-β1IFN-γ和IL-8 mRNA表达的影响 被引量:1

Effect of ruanganchengji on PBMN expression levels of TGF-β1 IFN-γ and IL-18 mRNA in patients with cirrhosis

摘要 目的:观察软肝冲剂治疗前后肝硬化(LC)患者外周血单核细胞(PBMC)中转化生长因子(TGF-β1)、干扰素-γ(IFN-γ)和白介素18(IL-18)的mRNA表达及分泌水平的变化。探讨软肝冲剂对肝硬化治疗的作用机制。方法:RT-PCR检测20例正常对照组和25例肝硬化患者软肝冲剂治疗前、治疗2、4、6月后PBMC中TGF-β1、IFN-γ和IL-8的mRNA表达水平;ELISA法检测PHA体外诱生PBMC培养上清中TGF-β1、IFN-γ和IL-18分泌水平。结果:肝硬化患者PBMC中的TGF-β1 mRNA表达水平及TGF-β1分泌水平显著高于对照组(P<0.01)。而IFN-γ和IL-18的mRNA表达水平及IFN-γ和IL-18分泌水平显著低于对照组(P<0.01)。经软肝冲剂治疗后,TGF-β1 mRNA表达水平及分泌水平逐渐下降,6个月后与治疗前相比有显著差异(P<0.01),但仍高于对照组;IFN-γ和IL-18的mRNA表达水平及IFN-γ和IL-18分泌水平逐渐上升,6个月后与治疗前相比有显著差异(P<0.01),但仍低于对照组。结论:软肝冲剂能通过下调PBMC中TGF-β1 mRNA和上调IFN-γ和IL-8 mRNA表达,对肝硬化有较好的疗效。 Objective:To study the changes of TGF-β1 mRNA, IFN-γ mRNA and IL-18mRNA in the peripheral blood mononuclear cells (PBMC) of patients with liver cirrhosis (LC) treated with Ruanganchengji. Method: 25 patients with liver cirrhosis were treated by Ruanganchengji. At different time (before therapy, and 2 months, 4 months and 6 months after therapy), The expression level of TGF-β1, IFN-γ,IL-18 mRNA in PBMC between LC group and control group (20) were measured by RT-PCR. The cytokines' Concentrations of TGF -β1, IFN-γ,IL-18 in PBMC in duced by phytohemagglutinin (PHA) were detected by ELISA. Result;The expression level of TGF-β1 mRNA in LC patients PBMC group was higher prominently than control PBMC group (P〈0. 001). but the expression level of IFN-γ, IL-18 mRNA in LC patients PBMC group was lower prominently than control PBMC group (P〈0. 001). After treated by Ruanganchengji, the expression level of TGF-β1 mRNA decreased gradually. It was lower prominently at 6 months than it was before therapy (P〈0. 001) . But it was still higher than that of control group(P〈0.011. The expression level of IFN-γ,IL-18 mRNA increased gradually. It was higher prominently at 6 months than it was before therapy(P〈0.01). But it was still lower than that of control group (P〈0. 01). Furthermore The Concentrations of TGF-β1, IFN-γ, IL-18 secreted by PBMC in duced by PHA also con firmed the same results. Conclusion: Ruanganchengji could treat Live cirrhosis through the decrease of the expres sion level of TGF-β1 mRNA and increase of that of IFN-γ,IL-18 mRNA in patients with Live cirrhosis.
