摘要
目的运用表面增强激光解吸附离子化飞行时间质谱(SELDI-TOF-MS)技术检测膝骨性关节炎患者关节液中蛋白质的差异改变,寻找骨性关节炎特异的生物标志,以期用于骨性关节炎的早期诊断。方法使用WCX2芯片,SELDI-TOF-MS技术检测20例膝骨性关节炎患者和8例正常对照关节液中蛋白质谱的改变。结果膝骨性关节炎患者与正常人比较,关节液中的蛋白质存在差异表达,共有6个蛋白表达水平发生变化,其中5754Da和15522Da蛋白峰仅在患者中表达,在正常人中不表达,13412.3Da蛋白符合Cys-tatin-c。结论SELDI蛋白芯片技术可以检测关节液标本,检测膝骨性关节炎患者关节液蛋白质的差异表达敏感性高,从而为骨性关节炎的早期诊断提供了一种新的方法。
[Objective] To detect different protein expression in synovial fluid between patients with knee osteoarthritis and healthy people using the SELDI ProteinChip platform ,in order to find the specific biomarkers which could be used for the early diagnosis of osteoarthritis. [Methods] Synovial fluid samples of 20 patients with knee osteoarthritis and 8 healthy people were tested by WCX2 chip on SELDI-TOF-MS Proteinchip reader. [Results] The protein expression in synovial fluid was different between patients with knee osteoarthritis and controls. Total 6 differently expressed proteins were detected, among which two peaks 5754Da and 15522Da were only expressed in osteoarthritis patients. In addition, the 13412.3Da peak was corresponded precisely to the molecular mass of Cystatin-c.[Conclusion] Synovial fluid could be applied on the SELDI Proteinchip platform, and using the SELDI technique to detect different protein expression between patients with knee osteoarthritis and controls was sensitive, this broadens a new applicable technique to diagnose osteoarthritis.
