黄酮甙类药物可能通过抑制核因子-κB信号通路改善Alzheimer病大鼠模型学习记忆障碍

摘要 目的:1,观察Alzheimer病(AD)模型大鼠核因子-κB(NF-κB)表达变化;2,研究黄酮甙类药物(葛根素)对AD模型大鼠NF-κB表达的影响。方法:wistar雄性大鼠18只,随机分为药物组、模型组、对照组,每组各6只。药物组和模型组双侧海马注射Aβ制成AD模型,对照组行假手术,双侧海马注射生理盐水。药物组按150mg/(kg.d)经腹腔注射葛根素治疗14d后处死;模型组和对照组给予3.75ml/(kg.d)生理盐水假治疗14d后处死。用Morris水迷宫评测学习记忆能力。westernblot分析和免疫组化两种方法检测大鼠海马NF-κB的表达,并作HE染色,观察病理改变。结果:模型组与对照组比较,平均逃避潜伏期延长,单位时间内跨越原平台次数减少,学习与空间记忆力受损(P<0.05)。葛根素治疗可使学习记忆力改善(P<0.05)。模型组大鼠NF-κB表达增强(P<0.01)。葛根素治疗可使过量表达的NF-κB水平下降(P<0.01)。结论:NF-κB可能参与了Aβ引发的AD大鼠模型的发病过程,黄酮甙类药物可能通过抑制NF-κB信号传导通路改善Alzheimer病模型大鼠学习记忆障碍。
