穿膜蛋白KAI1与intergrinα5β1、FAK在肺癌组织中的表达及其临床意义

摘要 目的本试验通过研究KAI1(Kang-ai-1)、intergrinα5β1和FAK基因编码的蛋白(focal adhesion kinase)产物在肺癌组织中的表达及其与临床病理学参数之间的关系,探讨它们在肺癌发生、发展、浸润、转移中的作用。方法应用免疫组化S-P方法检测非瘤肺组织、原发癌和转移癌中KAI1、intergrinα5β1和FAK蛋白的表达。结果在非瘤肺组织、肺癌组织和淋巴结转移癌中KAI1蛋白阳性表达率分别为100.0%,24.7%和0%,(P<0.01)。intergrinα5β1分别为0.0%,49.4%和83.3%(P<0.01),FAK分别为10.0%,48.3%和83.3%,(P<0.01),3个指标在三组之间的差异均具有统计学意义。KAI1和intergrinα5β1及FAK蛋白在肺癌组织中的表达都与患者的年龄、性别、肿瘤的大体类型无关,而与肿瘤的分化程度、临床分期密切相关。KAI1与intergrinα5β1呈显著负相关,intergrinα5β1与FAK呈显著正相关。结论KAI1与intergrinα5β1、FAK的异常表达与肺癌的浸润转移密切相关,检测这3项指标对预测肺癌的进展有一定的参考价值。 Objective To investigate the role of KAII, intergrin α5β1 and FAK proteins in occurrence and development and invasion and metastasis of lung cancer and their value in predicting the prognosis of lung cancer by analyzing the expression levels of KAI1 , in-tergrin α5β1 and FAK proteins in lung cancer and the relationship between them and clinicopathologieal parameters. Methods Lung cancer and normal lung tissue and metastasis cancer were deteeted and analyzed for their KAI1, and FAK protein level using immunobio-chemical method. Results The positive rate of KAI1 in normal tissue, primary cancer and metastasis cancer was respectively 100.0% , 24.7% and 0.0%. The positive rate of intergrinα5β1 was respectively 0.0% , 49.4% and 83.3%, The positive rate of FAK was respectively 10.0% ,48.3% and 83.3%. The difference of above three marker among three groups was significant (P〈0.01). The expressions of KAI1 , intergrin α5β1 and FAK protein in lung cancers had no relationship with ages and gender of the patients and the macroscopic type of cancer. But they significantly related with differentiated degree of tumor, clinical stage(P〈0.05). There is negative correlation between the expression of KAI1 and intergrinα5β1 protein, KAI1 and FAK. There is positive correlation between the expression of intergrinα5β1 and FAK. Conclusion The abnormal expression of the KAI1, intergrinα5β1 and FAK proteins may participate in the in-vasion and metastasis of lung cancer. Detecting their expression levels probably predict the malignant progression of lung cancer.
