摘要
目的将液态纳米组织工程骨注入兔实验性股骨头缺血性坏死区,观察其对股骨头缺血坏死的修复作用,并探讨其临床应用的可行性。方法在健康成年兔左侧股骨头建立骨缺损模形,获得成功存活动物54只,随机分成3组,每组18只。分别为A组:空白对照组;B组:注射单纯胶体纳米材料组;C组:注射共培养成骨细胞、内皮细胞复合胶体纳米材料组。A组不做其他治疗;B组将单纯纳米材料2 ml注入股骨头坏死区;C组将液态纳米组织工程骨2 ml注入股骨头坏死区。术后4、8、12周分批处死动物,每组每次处死6只。通过大体观察、X线摄片、光镜下组织病理学观察比较各组修复情况。结果影像学和组织学显示C组液态纳米组织工程骨组新生骨质量较高,骨愈合情况优于其他组。结论液态纳米组织工程骨有较强的诱导成骨能力,能有效地提高骨坏死病理条件下股骨头的骨修复能力,有望成为股骨头坏死治疗的一种手段。
Objective To observe the repairing effect of liquid nano-engineered bone on experimental avascular necrosis of femoral head in rabbits, and to evaluate the feasibility to use this material to repair osteonecrosis of the femoral head (ONFH) clinically. Methods Animal models of ONFH defect were established in 54 left femoral heads of healthy adult rabbits. They were divided into 3 groups randomly with 18 femoral heads in each groupgroup A: as the control group, group B: injection of NHAC only; group C: injection of both cells and NHAC. The specimens were harvested separately at the end of 4,8,12 weeks after operation (each time 6 rabbits in each group). Roentgenography and gross observation were performed, and histopathologieal changes were observed under light microscope. Results Group C had a better osteogenesis ability than group B and group A. Conclusion Liquid nano-engineered bone has a strong activity of osteoinduetion and can improve the ability of bone repair in the osteoneerosis areas of the femoral head. It may become one of the treatment modalities for femoral head necrosis in the future.
