NOS在颅内动脉瘤壁的表达

摘要 目的通过研究一氧化氮合成酶(NOS)在颅内动脉瘤壁的表达来探讨NOS对动脉瘤形成的影响。方法天坛医院2006年10月至2007年9月术中随机取得12例颅内动脉瘤,以同期5例脑肿瘤患者的颞浅动脉为对照,通过HE染色及Wester-blot蛋白分析法进行研究。结果HE染色显示相对于正常动脉壁,动脉瘤其内层、外层网架结构均有破坏;Western-blot结果NOS在动脉瘤中的蛋白表达明显增高。结论NOS在动脉瘤壁中的含量升高可能造成NO的积聚从而损伤动脉壁引起颅内动脉瘤的发生。 Objective To study the effect of NOS on the formation of intracranial aneurysms (IAs) by analyzing the expressive of NOS on the parietal of intracranial aneurysm. Methods Twelve tissue samples of intracranial aneurysms were taken randomly from Oct. 2006 to Sep. 2007 in Beijing Tiantan hospital, and five superficial temporal arteries samples taken from the patients with brain tumor in the same period were served as the control group. The pathological changes and the expression of NOS protein were analysed by HE staining and Western-blot respectively. Results HE staining indicated that the elastic fiber of aneurysmal wall, which plays a crucial role in the intact and stability of artery wall, was obviously destroyed when comparted with that of normal artery wall. The expression of NOS protein was significantly increased compared with that of the control group. Conclusion Increased NOS causes the accumulation of NO, which may induce the formtion of intracranial aneurysm by weakening the arterial vessel wall.
