离体筛选茄子抗羟脯氨酸突变体的初步研究 被引量:6

摘要 利用体细胞无性系变异筛选茄子的抗羟脯氨酸突变体,通过建立一套茄子离体培养技术,用浓度为8 mmol·L-1的羟脯氨酸(L-Hyp)作为筛选压力进行离体筛选,获得了茄子抗羟脯氨酸的愈伤组织,并再生培养成抗性植株。经耐冷性生理生化指标测定,抗性愈伤组织的脯氨酸含量与可溶性糖含量显著地高于对照,而丙二醛含量和电导百分率却显著低于对照,从而具有更好的耐冷性。再生植株的幼苗叶片脯氨酸含量显著高于对照,而电导百分率却显著低于对照,表现出更强的耐冷性。 This study aimed to use somaclonal variation to select hydroxyproline-resistant mutants of eggplant. Through establishing the in vitro culturing procedure of eggplant, 8 mmol·L^-1 L-hydroxyproline was used as selection stress to in vitro selection the mutants. Finally, hydroxyproline-resistant calli and their regenerated plants were obtained. The hydroxyproline - resistant calli had significantly higher proline content and higher soluble sugars, while significantly lower malondialdehyde content and lower electrical conductivity compared with controls when identified with physiological and biochemical indices of cold-tolerance of eggplant. The regenerated plants had significantly higher proline content and significantly lower electrical conductivity than controls. They also exhibited stronger cold tolerance than controls.
