摘要
目的观察莲房原花青素(Proanthocyanidins from Lotus Seedpod,LSPC)对高脂血症模型大鼠瘦素和肿瘤坏死因子(tumor necrosis factor-α,TNF-α)的影响。方法Wistar大鼠随机分成4组,即对照组(C组)、高脂血症模型组(H组)、LSPC高剂量组(HD组)、LSPC低剂量组(LD组),除对照组饲普通饲料外,其余各组喂饲高脂饲料并进行干预实验比较。4周后检测大鼠血脂,血清和心肌中瘦素及TNF-α的变化。结果与对照组相比较,H组血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)水平显著升高;血清中瘦素和TNF-α含量明显增高,心肌中TNF-α含量也明显增加,而心肌瘦素含量明显降低。HD组和LD组血清中瘦素和TNF-α含量明显降低,心肌中TNF-α含量也明显降低,而心肌瘦素含量明显升高,但对血脂水平无显著性影响。结论瘦素和TNF-α在高脂血症的病理和生理过程中起着重要作用,LSPC通过增加心肌对瘦素的调节敏感性,降低血清和心肌组织TNF-α含量,控制炎症反应,发挥对心血管系统的保护作用。
Objective To investigate the effects of Proanthocyanidins from Lotus Seedpod (LSP(2) on leptin and tumor necrosis factor-α (TNF-α) in hyperlipidemia model rats. Methods Wistar rats were randomly divided into 4 groups., control group, hyperliopidemia group, high dose of LSPC group and low dose of LSPC group. The rats were fed with high fat diet except control group and the intervention tests were carried out. After 4 weeks, blood lipid, leptin and TNF-α levels in serum and myocardium were assessed. Results Compared with control group, hyperlipoidemia rats had higher plasma leptin and TNF-α level, lower cardiac leptin level and higher cardiac TNF-α level. LSPC supplementation protected cardiovascular system by increasing cardiac leptin level. Accompanying these protective effects, LSPC decreased the level of leptin and TNF-α in plasma, and decreased the level of cardiac TNF-α. But LSPC had no significant effect on blood lipid level. Conclusion Leptin and TNF-α play important roles in the pathophysiology of hyperlipidemia. LSPC can increase cardiac leptin expression, decrease TNF-α contents in serum and cardiac tissue, control the reaction of inflammation, and have the anti-hypolipoidemia function.
