摘要
目的:探讨姜黄素对肺纤维化大鼠肺成纤维细胞增殖、凋亡的影响。方法:将体外培养的正常大鼠肺成纤维细胞(NLF)和肺纤维化大鼠肺成纤维细胞(MLF)分别于不同浓度的姜黄素中孵育;观察细胞生长状态变化;MTT法检测吸光值,计算细胞抑制率;流式细胞仪测定细胞凋亡率。结果:姜黄索对MLF有抑制作用,存在剂量依赖(r=0.723;P<0.01)与时间依赖性(r=0.625,P<0.05)。流式细胞分析显示,姜黄素处理组MLF凋亡率较阴性对照组(4.29%)显著升高(P<0.01),并呈药物浓度依赖性。姜黄素不能抑制NLF增殖,对其凋亡影响无显著性。结论:姜黄素在体外对MLF具有抑制增殖、诱导凋亡的作用;而对NLF无明显影响。提示姜黄素可以通过诱导肌成纤维细胞的凋亡而起到抗纤维化的作用。
Objective To investigate the effect of curcumin on lung fibreblasts of rats in both normal control group (NLF) and model group(MLF). Methods Fibroblasts were derived from lung tissue and were cultured in vitro. Then cells were treated with curcumin at different concentrations for different periods of time. The variations of morphology of pulmonary fibroblast were observed. Absorbance ratios of curcumin at different concentrations were determined by MTT assay. Apoptosis was quantitated by flow cytometry (FCM). Results The inhibition of curcumin on MLF was shown in both dose-dependent and time-dependent manner(P 〈 0.01 ), apoptosis rates were significantly higher than that of control group (P 〈 0.01 ). Curcumin can not inhibit proliferation and induce apoptosis in NLF. Conclusion In vitro curcumin can significantly inhibit proliferation and induce apoptosis of myofibroblast but not of fibroblast.
