Different expression of serum proteins in patients with hypertensive disorder complicating pregnancy

摘要 目的:分析妊娠期高血压疾病患者血清蛋白质谱,寻找妊娠期高血压疾病血清差异蛋白质,研究其在妊娠期高血压发病机制中的意义。方法:应用SELDI-TOF-MS技术检测31例血清标本(正常孕妇组10例,轻度子痫前期组12例,重度子痫前期组9例)的蛋白质质谱,用Biomark Wizard软件分析差异蛋白,并搜索鉴定蛋白。结果:筛选出6个质荷比(M/Z)为3263、3936、7979、7982、13773、15943的差异蛋白质峰,其蛋白质含量差异有显著性(P<0.05)。经蛋白质数据库搜索,与这6种差异蛋白质分子量最接近的蛋白质分别为血管活性肠肽、胰岛淀粉样多肽、泛醇-细胞色素C还原酶复合物7.8kDa蛋白、铁氧化还原样蛋白74个氨基酸、低诱导趋化因子A2前体、心房利钠因子前体。结论:SELDI-TOF-MS技术为研究妊娠期高血压疾病提供了可行的技术平台。 Objective To explore the serum protein mass spectrum in patients with hypertensive disorder complicating pregnancy to screen the differently expressed proteins. Methods 31 serum samples from 12 light eclampsism patients, 9 heavy eclampsism patients and 10 nomal pregnant women were bound to proein chip and tested by surface enhanced laser desoiption/ionization time of fligt-mass-spectrometry (SELDI-TOF-MS). The proteomic spectrum of all serum samples, searchde the differently expressed proteins were analyzed by Biomark Wizard software and obtained each differently expressed proteins accurately. At last, the differently expressed proteins were searched on the Uniprot protein database according to their molecular weight. Results At the different M/Z value range 6 proteins were obviously different in the patients with hypertensive disorder complicating pregnancy. Through searching database, these 6 proteins might be vasoactive intestinal peptide, islet amyloid polypeptide, Ubiquinol-cytochrome c reductase complex 7.8 kDa protein, ferredoxin-like, small-inducible cytokine A2 precursor and atrial natriuretic factor precursor. Conclusion SELDI-TOF- MS is a useful tool for the study of hypertensive disorder complicating pregnancy.
