摘要
目的:建立 RP-HPLC-ESI-MS 法测定大鼠血浆中欧前胡素的浓度,并探讨欧前胡素在大鼠体内的药代动力学。方法:采用 Shim-pack C_(18)色谱柱(5 μm,150 mm×2.0 mm),以乙腈-0.1%甲酸水溶液(75:25,v/v)为流动相等度洗脱,柱温为25℃,流速为0.2 mL·min^(-1),内标物为异欧前胡素。测定模式为选择离子监测(SIM mode,m/z 244),欧前胡素的保留时间为1.96 min,内标异欧前胡素为3.87 min。结果:在15~9000 ng·mL^(-1)浓度范围内,欧前胡素与内标异欧前胡素的峰面积比值与欧前胡素浓度有良好的线性关系。结论:该方法具有较高的灵敏度,能够准确、快速地测定血浆样品中欧前胡素的含量,可作为欧前胡素在火鼠体内的药动学的测定方法。
Objective:To establish an RP- HPLC -ESI -MS method to determine imperatorin in rat plasma and evaluate the pharmacokinetics of imperatorin in rat. Methods: The chromatographic separation was performed on a Shim - pack( VP - ODS) C18 analytical column(5 μm, 150 mm× 2.0 mm). The column temperature was 25 ℃. The mobile phase was the mixture of acetonitrile and 0. 1% formic acid solution (75 : 25, v/v) with the flow rate of 0. 2 mL· min^-1. Isoimperatorin was taken as the internal standard(IS). SIM( m/z 244)mode was selected to use. The retention time was 1.96 min for imperatorin and 3.87 min for IS. Results:In the range of concentration ( 15 -9000 ng · mL^-1 ) , there was a good linear relationship between peak area ratio of imperatorin/IS and concentration of imperatorin. Conclusion:The method with high sensitivity could rapidly and accurately determine the concentration of imperatorin in rat plasma, which can be used to study pharmacokinetics of imperatorin in rat.
