摘要
目的:建立三姐妹药材的 TLC 鉴别法以及药材中齐墩果酸和熊果酸的 HPLC-ELSD 含量测定法,考察广西不同产地、不同药用部位的药材质量。方法:以齐墩果酸为指标成分,三氯甲烷-甲醇(35:1)为展开剂,采用 TLC 法对三姐妹药材进行定性;以 Shim-pack VP-ODS(4.6 min×250 mm,5 μm)为色谱柱,甲醇-0.5%醋酸铵(85:15)为流动相,流速1.0 mL·min^(-1),Alltech ELSD 2000蒸发光散射检测器,采用 HPLC-ELSD 法对药材中齐墩果酸和熊果酸进行定量。结果:TLC 斑点分离清晰,简便易行;齐墩果酸和熊果酸线性范围分别为0.42~2.5μg(r=0.9998)和0.48~2.9μg(r=0.9997)。加样回收率齐墩果酸为98.3%(RSD=1.4%),熊果酸为100.3%(RSD=1.7%)。结论:本方法简便、准确,重复性好,专属性强,可用于该药材的质量控制和评价。
Objective : TLC and HPLC - ELSD methods were established to investigate the quality of Rabdosia temifolia( D. Don)Hara in Guangxi with different habitats and officinal parts. Methods :oleanolic acid as target constituent,Rabdosia temifolia( D. Don)Hara was identified by TLC ,used the chloroform-methanol(35:1 )as developing agent ; and the determination of oleanolic acid and ursolic acid were conducted by HPLC - ELSD with a Shim - pack VP - ODS (4. 6 mm × 250 mm,5 μm) column. The mobile phase was methanol - 0. 5 % ammonium acetate solution (85:15 ) ,at a flow rate of 1.0 mL · min^-1, and the detector was Alltech ELSD 2000. Results:The TLC method was easy and the spots were separated clearly;the calibration curves of oleanolic acid and ursolic acid were linear in range of 0. 42 - 2.5 μg ( r = 0. 9998 ) and 0. 48 - 2. 9μg ( r = 0. 9997 ), respectively. The average recoveries of oleanolic acid and ursolic acid were 98.28% ( RSD = 1.4% ) and 100. 3% ( RSD = 1.6% ), respectively. Conclusion :The developed method is simple, accurate, reproducible, specific and can be used for the quality control of Rabdosia temifolia ( D. Don) Hara.
