干扰素α-1b对裸鼠HepG-2瘤形成的抑制作用

摘要 目的比较干扰素α-1b(IFNα-1b)与聚乙二醇化干扰素α-1b(PEGIFNα-1b)抑制荷HepG-2裸鼠肿瘤形成的效果。方法取对数生长期的人肝癌HepG-2细胞接种于BALB/c雄性裸鼠背部皮下制备荷瘤动物模型。接种后14d按肿瘤大小将荷HepG-2瘤鼠随机分为溶剂对照组、PEGIFNα-1b组和IFNα-1b组3组,均连续5周皮下注射,每周1次。用电子数显卡尺测量肿瘤体积,第5周末全身麻醉处死裸鼠后测量体重、肿瘤体积和重量。结果观察期内PEGIFNα-1b和IFNα-1b对肿瘤的抑制率分别达56%和42%,未见其对荷瘤鼠体重有明显影响。结论PEGIFNα-1b对裸鼠体内HepG-2肿瘤生长有明显的抑制作用。 Objective To compare the effects of interferon alfa-1b(IFNα-1b) and pegylated IFNα-1b(PEG IFNα-1b) on inhibiting growth of human liver cancer cells in vivo.Methods Human liver cancer cells(HepG-2,2×l06/0.2ml) cultured in log phase were subcutaneously injected into the right rear flank of male BALB/C nude mice to establish the tumor model.After HepG-2 injection,the tumor growth was followed up for 14 days.The mice with HepG-2 tumor were randomly divided into placebo(n=7),PEG IFNα-1b(n=8) and IFNα-1b(n=8) groups.Mice received injection of placebo 0.2ml/mouse each time(three times a week),PEG IFNα-1b 150μg/mouse per injection(once a week) and IFNα-1b 50μg/mouse each time(three times a week),respectively,according to their grouping,and this regime lasted for five weeks.In order to evaluate anti-tumor activity,animals' body weight was measured every week,and the measurements of tumor volume and animals' body weight were continuously done till the mice were sacrificed at the end of the fifth week.Results PEG IFNα-1b and IFNα-1b induced a significant decrease in tumor volume and the animals' weight,the suppression rates onto tumor growth were 56%(2.190g vs.4.979g) and 42%(2.678g vs.4.979g),respectively.However,no significant difference was found in animals' weight among the three groups.Conclusion HepG-2(2×l06/0.2ml) injection into nude mice may induce a typical tumor model.PEG IFNα-1b(150μg/mouse per time,once a week) could significantly inhibit the growth of HepG-2 in vivo,and so does the IFNα-1b(50μg/mouse each time,three times per week).
