不同剂量胶原酶IV尾壳核内注射建立重度出血性脑卒中大鼠模型 被引量:1

Establishment of severe intracerebral hemorrhage model induced by injection of collagenase type Ⅳ with different doses into caudate putamen nuelei in rats

摘要 目的:通过胶原酶IV尾壳核内注射,探索建立一种稳定的重度出血性脑卒中大鼠模型。方法:45只SD大鼠随机分配到IV型胶原酶注射0.5U组(A组,15只)、0.75U组(B组,15只)及1.0U组(C组,15只)。制作脑出血模型后24h、72h,对大鼠进行神经功能学评分,检测肢体功能缺损情况。通过多媒体病理图像分析仪测定脑血肿体积大小,并通过HE染色观察组织病理学变化。结果:除A组1例外其余大鼠均形成明显血肿,C组形成血肿体积最大。3组大鼠均出现不同程度的神经功能缺损,以C组脑出血术后72h最明显。但C组术后72h病死率达73.33%,明显高于其他两组。24h、72h神经功能评分及血肿体积在A、B、C3组间作两两比较差异均有显著性(P<0.01)。脑血肿区组织病理学观察表现为典型的脑出血改变。结论:0.75UIV型胶原酶尾壳核内注射可以诱导建立稳定的重度出血性脑卒中大鼠模型,该模型肢体功能缺损明显,存活率高,能作为促进出血性脑卒中大鼠神经功能恢复研究的可靠应用平台。 Objective: To establish a reliable and severe intracerebral hemorrhage model in rats by injection of collagenase type Ⅳ into caudate putamen nuclei. Methods: A total of 45 SD rats were randomly divided into group A, group B and group C. Animals in 3 groups received injection of collagenase 0.5 U, 0.75 U and 1.0 U, respectively. The neurological behavior scores of rats were assessed at 24 h, 72 h after the injection. The volume of hematoma was detected by multimedia pathological imaging analysis system. Histopathological changes were observed with hematoxylin and eosin stain. Results: The experimental rats only except one in group A formed intracerebral hematoma, and group C showed the largest volume of hematoma. Rats in the three groups all showed neurological defect variously, and the most severe impairments was observed at 72 h after injection in group C. However, the mortality at 72 h after injection was 73.33% in group C,much higher than that in other groups. The scores of neurological behavior and volume of hematoma among group A, group B and group C showed obvious difference (P〈0.01). Conclusion: A reliable and severe intracerebral hemorrhage model in rats can be induced by injection of 0.75 U collagenase type Ⅳ into caudate putamen nuclei. The model has obviously neurological deficit and a high persistence. This is an useful and reliable method to study intracerebral hemorrhage.
